Pathogenic bacteria remain a major health problem because they are responsible for causing a large number of diseases. Their treatments with antibiotics lead to bacterial resistance that is increasing every minute. The main objective of this study was to prepare nanoparticles that would work as new therapeutic agents against pathogenic microbes. The nanoparticles were nanoemulsions (NE), produced by mixing the surfactants, soya phosphatidylcholine, polyoxyethylenglycerol trihydroxystearate 40 and sodium oleate, oil, cholesterol, and aqueous buffer, Tris-HCl. Three NE formulations (NEa, NEb and NEc) differed in fraction of surfactant and oil were formed. The droplet sizes of all of NE formulations were found in the range of 36-173.5 nm and their morphology were spherical and normally distributed, as determined by scanning electron microscope. Antimicrobial activities of the three NEs were examined against some Garm positive and negative bacteria, yeasts, fungi and virus. The highest antimicrobial activitiy was recorded against Staphylococcus aureus (+ve) and Enterobacter sp. (-ve). No activity was found against all tested yeasts or virus. However, NEs has fungicidal effect on some tested fungi. The mechanism of action of NEs was explored against Staphylococcus aureus and Enterobacter sp. The minimum inhibitory concentration (MIC) for St. aureus were 1.50, 1.25 and 1.25 ml for NEa, NEb and NEc, respectively whereas, MIC was 1.75 ml for all NEs in case of using Enterobacter sp. The effect of the three NEs on cell respiration, permeability, morphological changes and cell wall composition were also assessed using Staphylococcus aureus and Enterobacter sp. |
