Vitamin D (VD) is essential for overall health and wellbeing and it has been found to modulate the immune response. This study was done to evaluate the effects of different doses of VD on immunological and biochemical parameters and histology of some vital organs in rats. The total and differential white blood cells, red blood cells, and platelet counts; levels of hemoglobin, calcium, VD, antibodies (IgG, IgM, IgA, and IgE) were determined. Additionally, the food efficiency ratio, the body weight gains, the body and organs weights were determined. Thirty five adult male Wistar Albino rats, aged about 8 weeks, were randomly assigned to the following groups. The control group (8 rats) received 1 ml of distilled deionized water/rat for 22 days. The low dose group (LD, 9 rats) received VD at 1500 IU/rat, for 27 days (21 doses). The intermediate dose group (ID, 9 rats) received VD at a concentration 3,000 IU/rat for 28 days (22 doses). Finally, the high dose group (HD, 9 rats) received VD at 6,000 IU/rat for 28 days (22 doses). All doses were administered via oral gavage. Blood calcium and IgE levels increased for the ID and HD groups. There were significant decreases in WBCs RBCs, and platelets counts; and hemoglobin concentrations for the ID and HD groups. Body weight gains and food efficiency ratios were significantly decreased for the HD group and the liver, kidney and heart indices showed changes with VD. Other parameters were not significantly affected. The histopathology showed that the LD is safe while high doses result in focal degenerative changes in the liver and more so in the kidney. Lymphocytes were decreased in both spleen and thymus parenchyma. In conclusion, this study demonstrated that HD affects the immunological parameters pointing to harmful effect of hypervitaminosis D. |
